Confocal specifications
Leica STELLARIS 8 is a confocal point scanning system, including a White Light Laser, an Acousto Optical Beam Splitter (AOBS) and a highly sensitive, prism-based spectral detection design with computer controlled adjustable bandwidth for all internal detection channels.
The extended detection range up to 850 nm plus the expanded excitation range of the White Light Laser in the visible from 440 nm up to 790 nm allow the application and separation of an extended range of spectrally overlapping fluorophores (up to the NIR). The combination of Field-of-View (FOV) with Dynamic Signal Enhancement and LIGHTNING delivers superb image quality at full speed and in super-resolution.
In combination with the Power HyD detector family, STELLARIS 8 offers enhanced detection efficiency and photon counting. Including TauSense, STELLARIS offers a lifetime-based tool set that provides additional information in qualitative or semi-quantitative manner; using gate-based technology and mean average arrival time.
STELLARIS 8 White light laser (WLL)
- Allows a perfect match to peak excitation of fluorophores to ensure lowest laser exposure, optimal signal yield and emission separation of multi-labelled samples.
- Enables TauSense functionality: TauGating, TauContrast, TauSeparation, TauScan, GateScan.
- Allows recording of two-dimensional excitation-emission spectra supported by software for setup and display of 2D spectra.
- Pulsed excitation light source from 440 to 790 nm, up 8 single lines can be freely selected and used simultaneously.
- Freely tunable in steps of 1 nm.
- Laser power per line: 440 nm > 1.1 mW; 488 nm > 1.6 mW; 560 nm > 2.0 mW; 630 nm > 2.6 mW; 790 nm > 3.5 mW.
- Pulse frequency 78 MHZ.
- 8 channel AOTF (Acousto-Optical-Tunable-Filter) for rapid modulation of laser intensity, particularly for ROI (region-of-interest) illumination and excitation scans.

Power Hybrid Detector HyD S
All-round detector for high performance detection throughout the spectrum. Two times increased photon detection efficiency in the blue-green range compared to standard multi-alkaline photomultiplier. Leica Si-based technology.

Tandem scanning system for STELLARIS 8
Highest axial resolution with galvanometric scanner, switchable between resonant and non-resonant mode. Point scanning facilitates applications at high speed without compromising z-resolution or multi-parametric fluorescence as with other high-speed scanning systems.
Optical sections by point scanning, compatible with AOBS® and SP® detection.
Using resonant scanning, a higher fluorescence quantum yield and reduced photobleaching are achieved. Thanks to short pixel dwell times fewer molecular darks states are populated (anti-triplet synchronisation). In combination with Dynamic Signal Enhancement and LIGHTNING you achieve superb image quality at highest frame rates.

This high-speed raster scanning system is particularly well suited to live samples and physiological measurements, but it works just as well with fixed samples.
